Protective Effect of Leaf Extracts from Nelumbo nucifera on DNA Damage Against Oxidative Stress

Abstract

|
|
Objective: This study was carred out for the protective effect of leaf extracts from Nelumbo nucifera on DNA damage against oxidative stress
Methods: The 1,1- diphenyl-2-picrylhydrazyl (DPPH) radical and hydroxyl scavenging assay and Fe2+ chelating assay were performed for antioxidative effect and ¥õX-174 RF I DNA cleavage assay and intracellular DNA damage assay were used for inhibitory effect of intracellular DNA damage.
Results: The DPPH radical and hydroxyl scavenging activities and Fe2+ chelating activity were 90.82, 83.85, and 40.65% at 200 ug/ml. respectively. The similar pattern was found in hot-water extracts with a slightly lower activity. The inhibitory effect of intracelluar DNA damage was significant in both the Nelumbo nucifera extracts of ethylacetate and hot-water. Benzeneacetic acid, 4-Hdibenzoquinoline, Sarpagan-17-ol and Propenoic acid were identified by GC/MS analysis.
Conclusion: These results indicated that the leaf extracts of Nelumbo nucifera can be used as a natural antioxidants, which effectively inhibits the oxidative DNA damage.
